They are suitable for use with all common techniques, i.e. aqueous and solvent-based embedding media. In light sheet microscopy, the tissue pieces are illuminated through the lateral light entrance walls by means of a thin laser light sheet. The sample is moved vertically with respect to the light sheet in order to scan the sample in its entirety. The fluorescence of the individual sample planes is recorded from above with a camera and reconstructed in a 3D display software. There are two designs of sample holders for different sized tissue samples. They are made of optically high-quality and chemically resistant plastic with a glass-like refractive index of approx. 1.5. Both containers are flared at the top to facilitate filling and to move the rim menisci to the outside of the observation area. The latter allows distortion-free imaging through the liquid surface when using non-immersing dry objectives. The extended rim supports safe handling of the specimen holders and provides an area for a label or sticker for specimen identification. The specimen holders are stackable. The container offers usable space for specimens up to 40 x 15 x 10 mm. Its shape optimises the usable area for exposure and image capture with large dry objectives while consuming little embedding medium. The container is designed for specimen sizes that are fully imaged after clearing with one-sided illumination, such as mouse brains or other comparably sized tissues.
